REAL-TIME Cell Target Engagement

CellarisBio has rights to multiple patents protecting the MICRO-TAG™ cell target engagement technology. This robust enzyme complementation technology is based on split-RNase, enabling multiple unprecedented applications. 

Novel Fluorescence-Based Enzyme Complementation TechnologY

  • The small 15-amino acid small subunit of the reporter enzyme is linked to the target of interest. 
  • The large subunit is added externally to the reaction together with the FRET substrate. 
  • The complemented enzyme cleaves the FRET substrate and releases the fluorescent signal. 
  • Cellular target engagement is measured as fluorescence signal generated by FRET substrate cleavage. 
  • The level of signal is proportional to stabilization of the target by the tested ligand. 
  • The assay can be used with conventional T-agg method as well as Real-Time Step-Gradient method (see white paper below).
