- The large subunit is added externally to the reaction together with the FRET substrate.
- The complemented enzyme cleaves the FRET substrate and releases the fluorescent signal.
- Cellular target engagement is measured as fluorescence signal generated by FRET substrate cleavage.
- The level of signal is proportional to stabilization of the target by the tested ligand.
- The assay can be used with conventional T-agg method as well as Real-Time Step-Gradient method (see white paper below).
